Exocytosis: The Many Masters of the Exocyst
نویسندگان
چکیده
The exocyst is a conserved eight-subunit complex involved in the docking of exocytic vesicles. The exocyst has now been identified as an effector for five small GTPases, including Sec4, Rho1, Rho3, Cdc42 and, most recently, RalA.

Rho GTPase regulation of exocytosis in yeast is independent of GTP hydrolysis and polarization of the exocyst complex
Rho GTPases are important regulators of polarity in eukaryotic cells. In yeast they are involved in regulating the docking and fusion of secretory vesicles with the cell surface. Our analysis of a Rho3 mutant that is unable to interact with the Exo70 subunit of the exocyst reveals a normal polarization of the exocyst complex as well as other polarity markers. We also find that there is no redun...
متن کاملConservation and Specialization The Role of the Exocyst in Neuronal Exocytosis
The exocyst is an octameric complex mediating vesicle targeting and tethering at the plasma membrane for exocytosis. The role of exocyst in nervous system is unclear. In this issue of Neuron, Murthy et al. provide important insights: defects in the exocyst inhibit neurite outgrowth and neuromuscular junction formation; however, synaptic transmission persists.
متن کاملThe exocyst complex in polarized exocytosis.
The exocyst is an octameric protein complex, which mediates the tethering of post-Golgi secretory vesicles to the plasma membrane before exocytic fusion. The exocyst assembles by side-by-side packing of rod-shaped subunits composed of helical bundles. The targeting of secretory vesicles to the plasma membrane involves direct interactions of the exocyst with PI(4,5)P(2). In addition, a number of...
متن کاملThe exocyst is an effector for Sec4p, targeting secretory vesicles to sites of exocytosis.
Polarized secretion requires proper targeting of secretory vesicles to specific sites on the plasma membrane. Here we report that the exocyst complex plays a key role in vesicle targeting. Sec15p, an exocyst component, can associate with secretory vesicles and interact specifically with the rab GTPase, Sec4p, in its GTP-bound form. A chain of protein-protein interactions leads from Sec4p and Se...
متن کاملCharacterization of Mug33 reveals complementary roles for actin cable-dependent transport and exocyst regulators in fission yeast exocytosis
Although endocytosis and exocytosis have been extensively studied in budding yeast, there have been relatively few investigations of these complex processes in the fission yeast Schizosaccharomyces pombe. Here we identify and characterize fission yeast Mug33, a novel Tea1-interacting protein, and show that Mug33 is involved in exocytosis. Mug33 is a Sur7/PalI-family transmembrane protein that l...
